Roger Delgado was the first and a very good one as well. He died while filming Dr. Who and it contributed to friend Jon Pertwee (the 3rd Doctor) to leaving the show.

AND Eric Roberts (The Doctor Who movie on Fox)!
And there is also speculation that the Meddling Monk (Peter Butterworth) from the early years was actually the Master.
